Semaglutide

GLP-1 receptor agonistHuman clinical evidence

What is it?

Semaglutide is a peptide analogue that activates the GLP-1 receptor. It has been engineered for prolonged action compared with native GLP-1. Prescription products containing semaglutide have defined formulations and indications; a compound name by itself does not identify one of those products.

How it is studied

GLP-1 receptor activation affects glucose-dependent insulin secretion and pathways involved in food intake. Delayed gastric emptying is also part of its pharmacology. These actions differ from those of the dual agonist tirzepatide and the triple agonist retatrutide.

What the evidence shows

The official Wegovy product information summarises controlled trials of a specified semaglutide medicine and the populations studied. Clinical outcomes depend on the formulation, trial design and follow-up. Supplier strengths in this attachment represent vial contents, not the treatment regimens investigated in those trials.

Limitations & safety context

Labelled concerns include gastrointestinal effects, pancreatitis, gallbladder disease, dehydration-related kidney injury and a thyroid C-cell tumour warning based on animal findings. Other medicines and patient history affect risk. Semaglutide base, salt forms and independently labelled research materials must not be assumed interchangeable.
