PE-22-28

Spadin-related heptapeptideCell and mouse-model evidence

What is it?

PE-22-28 is a seven-amino-acid peptide derived from the spadin research programme. Its sequence is associated with a fragment of a sortilin-related propeptide. It has been investigated as an inhibitor of the TREK-1 potassium channel.

How it is studied

TREK-1 helps regulate electrical properties of cells. The foundational work compared shortened spadin analogues through channel recordings, neuronal experiments and mouse behavioural assays. Channel inhibition provides a mechanistic target, not proof of an antidepressant treatment.

What the evidence shows

A 2017 study identified the short active fragment and investigated its stability and behavioural effects in mice. Those endpoints support preclinical investigation. A mouse behavioural test cannot establish antidepressant efficacy, cognitive enhancement or appropriate use in a person.

Limitations & safety context

The cited study is not a human trial of PE10. Human benefit and safety remain unestablished by that paper, and the attachment supplies no clinical formulation data. Exact sequence and terminal modifications must match the experimental material when interpreting the results.
